چکیده

A murine monoclonal antibody specific for glycoprotein (GP)llla was prepared by immunization with a GPIIband GPIIla-enriched Triton X-1 14 extract of platelet membranes. This antibody, designated AP-3, was shown by indirect immunoprecipitation to react solely with GPIIIa derived from either P1 Alpositive or -negative individuals. The epitope on GPIIIa recognized by AP-3 is expressed on dissociated GPIIIa as well as on Ca 2-dependent complexes of GPIIb and GPIIIa, as shown by crossed immunoelectrophoresis in the presence or absence of EDTA. A previously described monoclonal antibody specific for the GPIIb/llla complex (AP-2) inhibited platelet aggregation induced by ADP, thrombin, collagen, or arachidonic acid (Pidard et al. J

Quantitation of membrane glycoprotein IIIa on intact human platelets using the monoclonal antibody, AP-3.

This antibody, designated AP-3, was shown by indirect immunoprecipitation to react solely with GPIIIa derived from either P1A1-positive or -negative individuals. Interaction of AP-2, a monoclonal antibody specific for the human platelet glycoprotein IIb-IIIa complex, with intact platelets.

A murine monoclonal antibody, designated AP-2, reacts specifically with the complex formed by human platelet membrane glycoproteins IIb and IIIa, but does not react at all with the individual glycoproteins. Purified AP-2 covalently coupled to Sepharose CL4B was used as an immunoadsorbent column to purify the IIb-IIIa complex from a preparation of Triton X-100-solubilized human platelet proteins...

Isolation and Structural Characterization of the Polypeptide Subunits of Membrane Glycoprotein lib-Illa From Human Platelets

We have previously demonstrated the isolation of platelet membrane glycoprotein lIb-Illa by affinity chromatography with a specific monoclonal antibody.1 We have now separated the polypeptide subunits lIb and lIla of the isolated glycoprotein by preparative sodium dodecyl sulfate polyacrylamide gel electrophoresis and have compared their structural features. Both llb and lIla contain -.1 5% car...

Calcium Ions and the Conformation of Glycoprotein lila That is Essential for Fibrinogen Binding to Platelets: Analysis by a New Monoclonal Anti-GP lila Antibody, TM83

Using a newly developed murine monoclonal antibody (MoAb). TM83. against glycoprotein lIla (GPIIIa) of human platelets, we have analyzed the relationship between platelet fibrinogen binding and conformational changes in GPIIIa under EDTA treatment. Crossed radioimmunoelectrophoresis demonstrated that TM83 reacted with only the GPIIb/lIla complex but also with GPllla alone. TM83 dosedependently ...

Temperature-dependent effects of EDTA on the membrane glycoprotein IIb-IIIa complex and platelet aggregability.

In agreement with previous studies, we observed that incubation of washed human platelets with EDTA at 37 degrees C for short periods caused an irreversible loss of their aggregation response to adenosine diphosphate and markedly diminished their capacity to bind fibrinogen. AP-2 is a monoclonal antibody that reacts with a determinant specific to the glycoprotein (GP) IIb-IIIa complex. We now r...
